Do I need to mirror the Sport Flex to put on an iron on T-shirt or have my Maker cut it the way it is written?
Iron-on vinyl or heat transfer vinyl should be mirrored or Flipped Horizontally whether you're using Silhouette or Cricut Design Space. The shiny plastic carrier sheet faces down on the cutting machine.

So this is true with any iron on or HTV vinyl in every case?
Everyday Iron-on is the one type of HTV that can be used as a base layer and then layer on top of it any other type of heat transfer material.
